It's actually Ms. BREX, :-).
How you set it up depends on what you wish to keep in the tank. Rinse the tank, and rinse your substrate. Place your substrate (gravel or sand) in the bottom of the tank, and then put in any rocks, corals, driftwood, etc., depending on the type of animals you intend to keep, and fill it halfway. Start installing the filtration, heater, thermometer, air pump, and lighting. For such a large tank you will want a very good filter. Finish filling the tank up, and plug in all of the equipment. Let the tank run for a few days to make sure the equipment is working correctly, and the temperature reaches the optimum level, then add SafeStart Plus or FilterActive and your first few fish.
